how long would it take to ride a moto (125) from pp to kampot or shv?

are there any roadblocks or checks on route?

240 km / 6 hours @ 40kph.

I did PP to Kampot in 2 hours before on a 150cc PCX. There was one road block/check, but no fine as I had a helmet and license/papers.

Yes, it can take one hour to get to the airport in a car or the bus during peak times.

2015, but I lived on the outskirts of PP so it was just jump on my scooter and go. Going from the city center can sometimes add 30 min to an hour or worse. Getting from Vietnam border to just outside of Phnom Penh last year (180 km), 2 1/2 hours. From the outskirts to the city center (5 km), another 1 hour and half.

The road down to Takeo is significantly less congested.

They may have fixed it because parts were rutted and washed out in 2014.

It takes about five or six hours if you ride defensively as you should. I have done it maybe ten or fifteen times.
