And "yes" - it's a real question. I passed this exam a couple of months ago, so here are some tricks:

1. Install the special application for your Android smartphone:

I'm pretty sure you can find the same app for iPhone as well.

2. It took me about couple weeks to remember all the questions and all the correct answers. You will find a lot of "high-level-true-Shakespeare's language" puzzles while training.
Please do not try to understand it, but remember the right answer. It will help you to pass the computer test (theoretical part of the exam).

3. Please do not think you are a good driver even if you have a lot of experience and have been driving a car for 30 years. It's Cambodia. I didn't see any drivers who managed to pass the driving test from the first trying. I saw some Chinese guys who had 10 or more attempts before they succeeded.
The right idea is to rent their old Camry (10 USD per half an hour) and get some practice right on "the playground" before your driving test. There are some cheats and tricks, but the guys you pay to (for renting a car) will help you to avoid the traps.

About the last question Q45 in the post above: I guess they mean "no one motorcycle rider can drive in the opposite direction."
I don't have any ideas about other questions - it's too sophisticated field of science.

New way of getting the Driving licence in Phnom Penh. Aeon mall II just opened a Week ago . Second floor. Staff very friendly. It was the same Price as the ministry of transport in Phnom Penh. So 10000 for the medical check and 30000 for the fee. So 40000 total. (10 dollars) . You need 2 photos. Old khmer licence and/or home Country licence (and One copy) . Original Passeport (and One copy) And proof of Residency (sangkat). For the payment they ask you to pay the 10000 cash and the 30000 with wing or Smart Luy ( the Smart office is not far from the main entrance on the ground floor. Fee 1000.) 45 minutes total. Great service.

I just got my license last week, I would add that you need a copy of the front and back of your current license, a copy of your visa, and that the Smart Luy is now located inside the the office itself.
